Determination of Bacteria: Staphylococcus Aureus and Staphylococcus Epidermidis
I am only picking between Staphylococcus aureus and Staphylococcus epidermidis. But, it was narrowed down to epidermidis due to some tests.
Each person work on an unknown bacteria. You’ll do Gram staining and biochemical tests over a few weeks. You will use your results from the various tests you do with your unknown to determine the genus and species of the unknown bacteria and then write a report Use complete sentences when you write the report. Do not use bullet points. This is a writing assignment and you will be graded on grammar, punctuation, and clarity of the writing. Format for the report 1.The cover page should have: Name Unknown number Genus and species of your unknown (10 points for right genus and species) 2. Write one or two paragraphs describing how you narrowed it down to one bacteria- starting with the Gram stain. Describe which tests were useful in eliminating some bacteria. Explain how the results of each of these tests were useful in narrowing down the list. Don’t just list the results and say what you think your bacteria is. You will be graded on the quality of the writing. Make sure your explanation is clear. Check for run on sentences, incomplete sentences, punctuation errors and grammar. Make sure the tense is consistent throughout the paragraphs. (10 points) 3. In the second column, list the appearance of your tube and whether it is positive or negative for each test. In a third column, list the interpretation/conclusion for each test. 4. In identifying the unknown bacterium labeled as number 26, an array of both staining and biochemical tests were meticulously conducted over several weeks. The initial step in the bacteriological identification was the Gram staining, which rendered a definitive purple coloration indicative of a Gram-positive bacterium. This preliminary classification was critical as it eliminated the possibility of Gram-negative genera, narrowing the scope of potential candidates.
Methodology and Test Results Interpretation
Following the Gram stain, the unknown showed a cocci morphology under microscopic observation, further limiting the possible bacteria range. A series of carbohydrate fermentation tests, including glucose and lactose, yielded positive results, suggesting the presence of enzymes capable of fermenting these sugars, characteristic of staphylococci, among other genera. However, the defining moment in the identification process was the negative result in the coagulase test. This characteristic is predominantly associated with Staphylococcus aureus, thus excluding it and pointing towards S. epidermidis, which is typically coagulase-negative. Nitrate reduction to nitrite, indicated by a positive nitrate reductase test, and the presence of catalase activity were consistent with the known metabolic pathways of S. epidermidis (Mainar & Leroy, 2015).
